Abstract:
Objective: To establish and evaluate the heart failure model induced by volume and pressure overload in New Zealand rabbits. Methods: Thirty-seven New Zealand rabbits were randomly divided into the heart failure group and the control group. The former group was subdivided into three groups. The volume overload group had rupture of the aortic valve, the pressure overload group was created by a suprarenal stenosis of 50% of the abdominal aorta, and the combination group underwent both operations. Control group only had dissection of arteries. Hemodynamic and echocardiographic characteristics and BNP level were evaluated 8 weeks after the operation. Results: Hemodynamic results showed that heart rate, aortic systolic pressure, aortic diastolic pressure, pulse pressure, left ventricular end systolic pressure and left ventricular end diastolic pressure were significantly higher in the combination group than in the other groups (all P<0.05). Echocardiographic results indicated that left ventricular end diastolic diameter, left ventricular end systolic diameter, interventricular septum thickness, left ventricular posterior wall thickness, fraction shortening (FS), and E/A ratio were significantly higher in the combination group than in the control group (P<0.05) , and EF was significantly lower than in the other groups (all P<0.05). BNP concentration of rabbits in the combination group significantly increased compared with that in the other groups (all P<0.05). Heart weight /body weight and lung weight/body weight in the combination group were significantly higher than those in the other groups (all P<0.05). Conclusion: The animal heart failure model induced by volume and pressure overload can be easily established. Pressure overload and volume overload have different effects on the cardiac function.
